Glycoproteins: Liquid Chromatography

چکیده

Many proteins in cells and biological Suids are glycosylated, and these glycoproteins are present in animals, plants, microorganisms and viruses. The most commonly occurring monosaccharides found in oligosaccharide attachments to mammalian proteins are D-mannose (Man), D-galactose (Gal), D-glucose (Glu), L-fucose (Fuc), N-acetylglucosamine (GlcNAc), N-acetylgalactosamine (GalNAc), and Nacetylneuraminic acid (sialic acid or NeuAc). The primary structure of glycoprotein glycans and their biological functions have been gradually unravelled by the improvements of methods for isolation and structure determination. High performance liquid chromatography (HPLC) is one of the most commonly used methods for the isolation and analysis of both glycoproteins and their derived carbohydrates, mainly due to the excellent resolution, ease of use, the generally high recoveries, the excellent reproducibility of repetitive separations, and the high productivity in terms of cost parameters.

Identification, Isolation, and N-Terminal Sequencing of Style Glycoproteins Associated with Self-lncompatibility in Nicotiana alata

S-Gene-associated glycoproteins (S-glycoproteins) from styles of Nicotiana alata, identified by non-equilibrium twodimensional electrophoresis, were purified by cation exchange fast protein liquid chromatography with yields of 0.5 to 8 micrograms of protein per style, depending on the S-genotype of the plant. The method relies on the highly basic nature of the S-glycoproteins. The elution profi...

Purification and Partial Amino Acid Sequence of Human Platelet Membrane Glycoproteins tIb and lila

The glycoprotein (GP) llb-Illa complex was isolated from human platelet membranes by immunoaffinity chromatography using a monocbonal antibody specific for GP bIb-lIla. GP llb and lIla were further separated in the presence of sodium dodecyl sulfate (SDS) by gel filtration high-performance liquid chromatography (HPLC). Two cycles of this procedure yielded almost complete separation of homogeneo...
